Just add "/Developer/usr/bin" into "binpath" in /opt/local/etc/macports/macports.conf
I have the same problem too after upgrade to snow leopard and install a new XCode for iPhone 3.1.

I assume you unchecked the checkbox "UNIX Development Support" when you installed Xcode / iPhone SDK? If so, go back and install it with that option checked. The MacPorts installation instructions specifically mention this necessity:
Section 2.2.4:
Ensure that the option for command line development, which is labeled Unix Development in the Xcode 3.x installer, is checked.

How & when did you install MacPorts? What version of MacPorts, the OS, and Xcode?
make
should be/usr/bin/make
