Description
I'm running 10.12 public beta 5 (16A294a) and have installed version 8.0 (16 Aug 2016) of the command-line tools. I cleared out all my ports with "sudo port -f uninstall installed && sudo rm -rf /opt/local/var/macports/build/*", installed MacPorts from source, and did "sudo port selfupdate && sudo port upgrade outdated" prior to attempting to install glib2.

glib2 2.48.2 installs fine on macOS Sierra beta 5. Make sure you install and xcode-select
Xcode 8 beta or later, and install the corresponding version of the command line tools. Earlier Xcode and command line tools versions cannot be used with Sierra.

Software update had me on Sierra beta 6, command-line tools version 8.0, but Xcode 7.3. Installing and selecting Xcode 8 beta gave me a clean compile and install of glib2 2.48.2_0.
